ET-743

Background

Dr. von Mehren has written an excellent introduction to for this agent called, "ET-743: A Review of Recent Results".

Research Results: Ewing's Sarcoma

  • 2002 ASCO phase 2 results which mention 2/8 ES success; authors are in Italy and Switzerland

  • Closed COG pediatric phase 1 trial (Baruchel at Toronto was the lead)

    • 2002 ASCO abstract on this study discussing dosing

    • 1/1 response for Ewing's. There was prolonged partial response for a relapsed ES patient on this trial at dose 1100 mg/m2; the patient received 16 treatments (one year) and was still responding when she stopped the drug. more information

  • 2002 abstract stating that ET-743 kills Ewing's sarcoma cells. Both drug-sensitive and drug-resistant bone tumor cells were used (note: this is cell line research)

Research Results: Non-Ewing's Sarcoma

  • On the Phase 1 ET-743+Doxil trial at Fox Chase, 2/2 sarcomas have become stable; results not published yet

  • 2002 ASCO phase 2 results, giving 24% partial success rate for soft tissue sarcomas failing prior chemo; authors are from Memorial Sloan-Kettering Cancer Center and Dana Farber Cancer Institute

  • Closed phase 2 trial in Europe (Ewing's sarcoma NOT allowed)

  • Closed pediatric phase 2 trial at Memorial Sloan-Kettering Cancer Centefor osteosarcoma

    •  2002 abstract on study; it didn't work well in the "heavily pretreated osteosarcoma population" studied

  • 2002 abstract discussing phase 2 trial results; authors are from Dana Farber Cancer Institute and France; no discussion on efficacy
